Her Grandfather is the Great Hashirama Senju. Her Grandmother is Mito Uzamaki, the former jinchuuriki of the Kyuubi. She is atleast 1/4 Senju and 1/4 Uzamaki, hailing from strongest clans of the Narutoverse.

Her Senju DNA gives her a strong body, her Uzamaki DNA gives her extremely high chakra levels and a very powerful life force.

Who would have thought that the Naruto character with the best Genetics is Tsunade of all people?! So why isn't she more powerful? hmmmm....

The Namikaze Clan is nowhere near the caliber of both the Uzamaki and Senju.

Also since the Uzamaki are considered to be descendents of the Senju, Tsunade is likely more than 25% Senju, possibly 30-40% even. More than any other living shinobi...

Well imo it's because Tsunade probably wasn't all that interested in being a shinobi when she was young so she never really developed an interest in fighting like some other characters. Remember, she was afraid of blood so I don't think she was too fond of fighting. Also after she became a Sannin she left Konoha and started to do nothing but gamble.

Other strong characters like Madara, Itachi, and Hashirama? They were kicking ass before their teens and didn't do much else since.
