No, it's not that he couldn't read it, he just didn't need to read it.

The purpose of going to the Naka Shrine was because the Uchiha Tablet was needed to unseal the Shinigami. That is why Sasuke referred to the Tablet as "the important thing downstairs"

The Tablet was created by Rikudou Sennin, and the Shiki Fuujin was created by him as well.

Well Obito said that if the Tablet is read in the order of MS, EMS and rinnegan we get to know the secrets of the SO6P and Sasuke read the tablet which was before leaving the village so he just knew about the power of MS to control Kuruma.

So if he would got a chance to further read it with EMS he would have came to know about the origins and the links between SO6P senju's and the Uchiha's as well. But he didn't had EMS at that time so he didn't knew about it.
