I wanted to say that, Madara manipulated Hashirama by using his EMS powers (which are still unknown in the manga). Madara prepared Hashi's death by killing him with the poison since Madara knows much clearly about Hashi's healing ability that no one can penetrate through.
I wanted to make the things clear with the facts we heard until chapter 626 and then what he learned from Hashi's last words in 626. That made me think what might had happened at the Valley of End and what if of all the places Madara used his EMS powers to finish off Hashi slowly but too badly.
