I think it's highly plausible that Mito helped Hashirama during his fight with Madara by binding and sealing Kurama, take a look at this:

This is the Viz Translation of Kushina's statement, it says "during that battle" which probably implies Mito's presence during that fight. She sealed Kurama within herself.
Since Kishimoto loves making parallels, this would mirror Minato's fight with Kurama wherein Kushina bound the Kyuubi with her Uzumaki chakra chains.

The possibility of Mito sealing Kurama during that fight is further strengthened by the fact that the final phase of the battle happened in the Valley of the End

And I don't think neither the full grown Kurama nor the Mokuton summon would fit at such a cramped location, the waterfalls is all natural so I doubt that particular location was as spacious enough even then as to accommodate the 1K hands summon or the Full Grown Kyuubi

So either one (Kurama and 1k hands summon) got de-summoned or got bounded during the fight because neither would fit in VotE.
Not to mention Kurama was covered in Susanoo Armor that would've prevented Mokuton from binding it

So the possible scenario that could've occurred was:
a.) Madara w/ Kurama destroyed the Mokuton construct and Mito arrived to save Hashirama by sealing Kurama (mirroring Kushina aiding Minato)
Or
b.) Hashirama took Kurama from Madara's control but still had to chase after him to VotE so Mito lent a helping hand so Hashirama could continue with the chase.
This could also explain the apparent misogynist attitude Madara has (eg. Tsunade being a weak woman) it must have been shameful for him and hid the fact that a woman was instrumental in his defeat.
