I think the true power of Mokuton, is that it allows the user to absorb Natural Energy. I think Mokuton much like Jugo's clan is a gateway to SM. I think the true power of Senju was in mastering and using SM. The reason everyone was after Hashirama was, because his DNA (like Jugo's) provided an easy gateway to SM.
We have seen how SM gives greater durability and heals people. Jugo is able to heal himself and so is Hashirama.
Failing to control SM has disastrous consequences. Frog SM turns you into a stone, it seems Snake SM turns you into a snake, Jugo's clan makes you go crazy and perhaps Hashirama's turns you into a tree.
When Kabuto used SM it made his snakes grow a lot bigger. Look at the size of the Trees Hashirama creates. It is probably the Natural Energy, which allows them to grow to this incredible size.
Conclusion
Just like the Uchiha could gain greater power through the MS, the Senju could create greater power through SM.
Hashirama by a freak chance, like Jugo, had DNA that constantly drew in Natural Energy. This was part of the reason why he could grow such huge forest, had incredible stamina and could heal himself.
Hence this is the reason why all the wannabe RS are after Hashirama's DNA specifically. Since it is his genes, which allow Natural Energy to freely be absorbed. In the case of other Senju they have to train.
This ties in Hashirama, with the Younger Son and SM with the Senju.
