Madara held a grudge against the Senju-Uchiha alliance at the time of the creation on Konoha.
He was the only Uchiha who rebelled against it. The reason is most likely that he'll lose his father in the upcoming battle and this will break the bonds b/w Hashirama and Madara.

The way things are right now in the Chapter, Madara and Hashirama are really good friends who are ready to oppose their clans just to save each other. Now with the upcoming fight it is likely that Madara will lose his father.
I'm not sure that Hashirama will lose his father too but I'm sure about Madara.

This even will turn both Madara and Izuna completely against the Senju clan. That's when they will awaken MS.
During war it is likely that Izuna saves Madara the same way Obito saved Kakashi and then the cycle of hatred will start once again.

I've thought of a good storyline from this point:

The Uchiha and Senju father and son duo will fight and the battle will turn out to be a draw.
The Senju play dirty and commence a wide range attack aimed directly at Madara. Madara's father gives up his life to save Madara. Madara stands with mixed feelings of mistrust and despair.
Izuna then forces Madara to retreat.

I really do believe they will awaken MS when losing the father against the senju. That will force Madara to decide between his clan and his friend. Since he holds his little brother dear and he means the world to him, he will decide against Hashirama. Fully understandable.

I think the events after will show how sad that story can be. Madara and Izuna will train together as it was shown before. Izuna will pretty much take Hashirama's role in his life. Their bond will grow, the bond with Hashi will break.

Tobirama will most likely show a massive grudge against the Uchiha (maybe his father will die as well). And I think he will be responsible for Izuna's death as a blind man. Then Madara will go fully insane and want revenge for his brother. That's how a good guy broke and how a bad guy was born.

All the events after is what we already know. Madara is a truly tragic figure and deserves nothing but a good and peaceful ending.

That's a view that gets stronger and stronger with each and every chapter, as much as some people still disagree and love to see him as a bad guy who will never forgive. I can only hope and see how things go. And I so want him to see Hashi again.
