In these flashbacks, we can gauge young Madara and Hashirama were people who wished for true peace. I think their friendship really bonded in their early years and they made a vow to find peace together.
I am suspecting Madara and Hashirama devised a plan to conquer every possible clans, and unite them under Uchiha and Senju.
Something like -
Madara: I will conquer half the existing clans!
Hashirama: I will conquer the other half too!
Hashi + Madara: We will then unite them as one!!!
They succeeded in doing so as their clan names were truly feared in that era.
I guess it was during their peak, they decided to unite all the shinobi clans as one and they had to choose a leader. Somehow, Hashirama was picked ahead of Madara and this trigger Madara to declare Hashirama as his eternal rival. Probably Madara felt he was the older boy and deserves to be the overall leader.
It was not meant to be.
