I have come to the conclusion that based on the evidences presented in the manga up to the present point that the destruction of Uzushiogakure was instigated none other than by Madara himself through his agents: the shinobis of the Kirigakure, for the expressed purposes of eliminating a potential threat, and most importantly, the acquisition of the host of the Rinnegan: Nagato Uzumaki.
Now, based on Kushina's statement to Naruto, she was aware of the destruction of Uzushiogakure, which means the destruction had occur before or within her own lifespan:
And based on her statements also, it is safe to conclude that the destruction occur immediately before or after Kushina's arrival in Konoha, since the transplantation of the Rinnegan to Nagato occured approximately 32 years BSS, and Nagato only awakened it in 28 BSS when he and his family were already living in Amegakure after Uzushiogakure's destruction, the same year when Kushina arrived in Konoha, which makes the destruction of Uzushiogakure within the time frame of Kushina's childhood.
Now, concerning the Second Shinobi World War, the participants were Konohagakure, led by the Third Hokage, Sunagakure, likely led by the Third Kazekage, Amegakure, led by Hanzō of the Salamander, and Iwagakure, presumably led by the Third Tsuchikage.
So the participants were:
1.Konohagakure
2.Sunagakure,
3.Amegakure
4.Iwagakure
But wait, aren't there FIVE shinobi villages? of the Five Great Shinobi Countries? but which one is missing:
Answer: Kirigakure, "Village Hidden by Mist"
So why were they missing? why would one of the five great shinobi villages not participate in a shinobi world war? what were they doing at the time?
The Answer: The Destruction of Uzushiogakure
It was the perfect timing, Konoha, the ally of Uzushiogakure, was engaged in a war with the other three villages, Kirigakure under the guise of being neutral, could muster its' military force and focus on one objective, the destruction of the Hidden Leaf's village ally: Uzushiogakure
Do you not wonder why they only chose to participate in the Third Shinobi World War, and even so only near its' climax? it was because they had ulterior motives in not joining in the war, the destruction of the Uzumaki village and the scattering of the Uzumaki Clan members.
But who was in control of Kirigakure during that time?
The Answer: Madara Uchiha
We all know that Yagura, the fourth Mizukage was being controlled by Tobi, but, if my suspicions are correct, Tobi only inherited control of Kirigakure when Madara died meaning Madara most probably controlled Kirigakure since before or during the Second Shinobi World War.
[This also leads me to believe that Kirigakure, upon the orders/control of Madara, was the one who staged the death of Rin in order to gain alliance from Obito, who became Madara's protege/successor]
But why would Madara want to destroy Uzushiogakure? well since the Uzumaki Clan possessed very powerful Fuinjutsu (and are even rumored to be Rikudou Sennin's original bloodline) then it's logical to assume that Uzushiogakure would pose a very serious threat to the Eye of the Moon Plan
They were a threat and they needed to be eliminated at any cost, or the very least, scatter them in the wind like a thousand broken glasses.
And since a herd divided is easier to manipulate/control than one that is united, then it would have been easier for Madara to obtain his prized possession, his other motive:
Madara needed him to further awaken the Rinnegan's powers, esp Rinnei Tensei which would've been used to revive Madara when the Eye of the Moon Plan commences:
With the destruction of Uzushiogakure, Madara had achieved two goals:
1. Eliminate the Uzumaki threat
2. Acquire an ample host for the Rinnegan
The Eye of the Moon Plan commences, a plan painted with the blood of the Uzumakis.
