ok I know this is a lot so i apologize for the wall im just trying to cover the bases
well danzos arm was said to have been influenced by orochimaru, and with all this use of senju dna is it a stretch to say that oro has been searching his whole life for a body capable of mokuton, and could that be why he thinks sasuke has the perfect body. I think madara has proven that uchiha take to senju dna pretty well unlike most, i mean yamato was the lone survivor of oros experiments, yet both obito and madara wield mokuton no prob.
Edo madara also proved that kabuto and oro know, that the sharingan progresses to rinnegan, and that senju DNA is the key as the sage is the combination of senju and uchiha. For how long have they known this though?
We know oro was obsessed with jutsu and wanted every jutsu, and thats why i think his plan was not just to obtain a sharingan, but a rinnegan as well. that is i think the only way to master all jutsu of all natures as nagato was able too. which is only possible if he finds the perfect body, an uchiha body.
When oro joined akatsuki i think he saw pain the leader and wanted his eyes, the eyes of the sage father of both senju and uchiha, which is how he hypothesized that the rinnegan was the product of both senju and uchiha. At this point oro has been kicked out of the leaf for his yamato/shodai experiments, which he thought had all failed, so he gets this new idea i will steal the perfect body for senju dna and gain the eyes of the sage in one go, i will steal the body of a pure uchiha, uchiha itachi. Oro attacks itachi fails and is dissmissed, but oro is not the type to give up so easily.
so now oro has left akatsuki, but he stays in contact with danzo, who has recently gained an absurd amount of sharingan including uchiha shisui. danzo then goes to oro, who is overjoyed at the thought of combining his senju dna with sharingan, and while danzo will consider izanagi a success its not what oro wants. So oro keeps searching, the nakano shrine, uzumaki ruins, until he finds juugo and is lead to the ryuchidou.
the curse mark stems from juugos power, which always went beserk, and we all know the answer to controlling a power that you cant normally control lol.senju dna, exactly like danzos arm. If you recall when juugo took a piece of kabuto he said that is what his transformation was like before the curse seal. not to mention juugos ability is similar to mokuton as his body can grow and shrink like a living mokuton infused with sage chakra, i think senju dna may have an influence on juugos transformation, oro thought he found his first compatible host for senju dna.
now i think that at the time oro had given sasuke the curse mark, oro had already hypothesized about the rinnegan, whether sasuke came first or madara i think its hard to say. but i think sasuke was given a dose of senju dna with the curse mark, so that sasuke wouldnt be devoured by the curse mark and go berserk, and more importantly to give sasuke hashirama dna(mokuton) in preparation of oro taking over his body and eventually gaining rinnegan.
