No, there are ways to unlock the MS besides killing your best friend....Itachi was only trying to push Sasuke in to gaining the MS powers so he could fight him and die and Itachi didn't give Sasuke his MS powers because if that was the case then Sasuke should also have the same MS design as Itachi and should have a God-like Tsukuyomi at Itachi's level, but that's not the case.

1.Either throw killing a best friend.
