Agreed, there is no way of knowing if the DNA was Karin's; who knows what sort of stuff an Uzumaki Lady keeps in her desk. And---as has already been mentioned---Sakura's posture and the hand over her belly in "The World to Come" on panels 9 and 10 suggest that she might have been pregnant...
