Which is best and which combo would you choose?

I'd choose Sage Mode and Mokuton cause GG, but Sage Mode and Shikotsumyaku seems like it'd be Hashirama level.. Jk SS too big

The best powers are Perfect Jinchuuriki or Eternal Mangekyou Sharingan.

Mokuton and Sage Mode are also just as great but conditional. Mokuton's power depends upon the user's chakra. And Sage Mode's power depends upon the user's abilities. So if you have a powerful chakra and life force, then Mokuton is also a good choice. And if you have top notch abilities, then Sage Mode empowerment is beast.

But in general, EMS and Perfect Jin are powers with standard OP power level so they're easy to decide and select.

The best combo would probably be EMS+SM for me.

Lol "If you are as strong as Madara" obviously encompasses the entire statement, so why add "if you are as strong as Hashirama" there? :lol

And no, SM+EMS is nowhere near SM and Mokuton if given to the same exact user. Madara has the potential to replicate all of Hashirama's Mokuton at the same level. SM EMS Madara only has a PS w/ Senjutsu while SM Madara w/ Mokuton has Shinsuusenju, which dumps on PS regardless of Senjutsu.

Not sure what you are even talking about with your first post when almost every power depends on the user's strength bar Jins, since the bulk of their power comes from the Bijuu.
